You do this by creating contexts, which can be locations or specific situations you'll find yourself in with your computer. It doesn't have to revolve around a specific location. You can think of it more as a situation during which you'd want certain settings to be active. This may be "music listening time" or "visiting family" instead of just "work" and "home." Research carried out at Boston College, in collaboration with scientists at MIT and the University of Oxford, has led to the development of an efficient and highly selective catalyst for ring-closing olefin metathesis, one of the most widely used reactions in chemical synthesis, the team reports in this week's issue of the journal Nature.
The team used the new catalyst, part of a large and important class of carbon-carbon double bonds, to synthesize epothilone C and nakadomarin A, both of which are molecules that have been shown to be potent anti-cancer agents. Epothilone A, an equally active agent can be prepared from epothilone C.
"Catalytic ring-closing olefin metathesis is probably the most widely used chemistry method to access biologically active molecules," said the report's principal author, Amir Hoveyda, the Joseph T. and Patricia Vanderslice Millennium Professor of Chemistry at Boston College. "The catalyst that we have identified as optimal in this study is remarkably efficient and selective. Considering the demonstrated significance of this class of reactions, even though it has thus far been largely non-selective, findings indicate that our catalysts and method will have a major influence on the future of chemical synthesis."
Carbon?carbon double bonds, also referred to as alkenes or olefins, are present in many medicinally relevant and biologically active molecules. Hoveyda and MIT Professor Richard Schrock, a co-author of the Nature report, have collaborated since 1997 in developing new and effective catalysts for such transformations. Schrock shared the 2005 Nobel Prize in Chemistry for discovering one of the earliest types of olefin metathesis catalysts.
Alkenes exist as either the zigzag shaped trans olefin, or the E isomer, while others take the "U" shape of the cis olefin, or the higher-energy Z isomer. Z isomers require a catalyst that must be sufficiently active to be capable of promoting the chemical reaction while maintaining the cis olefin's U-shape geometry. Preserving both characteristics in a catalyst leads to reactions that deliver Z alkenes, which can be found in a large number of medicinally significant molecules and serve as starting materials for some of the most commonly used transformations in chemistry.
Ring-closing metathesis allows the formation of any ring size from readily available linear chains. However, the process is typically non-efficient or results in the formation of a significant amount of the undesired isomer, which often has less or no biological activity.
Using tungsten, the team found a catalyst that offers exceptional reactivity and selectivity while being sufficiently stable to air and moisture to make its use particularly attractive, said Hoveyda, who with Schrock co-founded the Swiss firm XiMo, which has licensed the new catalyst and its technology.
To demonstrate the utility of their catalysts, the team focused on epothilone C and nakadomarin A. Several other leading research groups in the world have previously prepared these biologically active natural products, but, in all cases, the critical ring closure step has been non-selective. This lack of selectivity badly damages the overall efficiency required to prepare these much sought after molecules, since ring formation comes at the tail end of a long sequence of reactions, sometimes as many as 20 steps.
"Losing half of your materials after you have spent so much time and energy and thought preparing it is nothing short of devastating, particularly when you cannot even separate the two isomers," Hoveyda says.
Epothilone C can be produced through fermentation, but creating various analogues by the same method would be extremely difficult. Nakadomarin A, found in marine environments, is only available in minute quantities. The highly selective process developed by the team will allow scientists to access significant quantities of these natural product as well as many of their analogs, which might prove to have more attractive therapeutic attributes.
The findings are the most recent from 14 years of collaboration between the Hoveyda and Schrock labs, work that has been supported by the National Institutes of Health since 1999. Also contributing to the study were Miao Yu and Chenbo Wang of Boston College and Andrew F. Kyle, Pavol Jakubec and Darren J. Dixon of the University of Oxford (UK).

NEW YORK?? A 47-year-old Manhattan mother was fighting for her life at Harlem Hospital on Wednesday, two days after she was struck by a shopping cart that was thrown from four stories above her at East River Plaza Mall in East Harlem.

According to police, the boys were playing around on the fourth-floor walkway of the mall when they decided to push shopping carts against the railing, New York's WNBC reported. One of the carts fell four stories down on top of Marion Salmon Hedges, who had been at the mall to shop for Halloween candy with her 13-year-old son.
The 12-year-old boys accused of throwing the shopping cart were arrested Tuesday and charged with juvenile assault. The two, who were described by neighbors as coming from underprivileged families, were turned-in by a 14-year-old friend who had tried to stop them, the New York Daily News reported.
According the NBC News' Stephanie Gosk, Hedges' husband Michael Hedges said on his facebook page that he hopes the two boys get the kind of care they need to become responsible members of the community.
"She's going to be, in the best of all cases, in rehabilitation for months," the newspaper quoted Michael Hedges as saying on Tuesday.
"She's under sedation. She's intubated. She has a thousand machines hooked up to her. It?s very disturbing," he reportedly added.

Hedges, a real estate agent who lives on the Upper West Side, is known for her charity work, WNBC reported. She was a board member of the Samuel M. Isaacs community center and had a special interest in the youth programs. Hedges is active in her church, the Daily news reported.
"That's somebody's mother, someone's daughter," said shopper Monique Johnson, according to WNBC. "Whoever did it should be prosecuted to the fullest extent."
Director Wanda Wooton told NBC New York the situation was "unbelievable," "especially because it happened to Marion who is so giving and wonderful."
"It's horrendous," said Angelica Gonzalez, a shopper whose friend saw the aftermath of the incident. "It's horrible what these kids do."
The pranksters were charged in family court with assault as juveniles. Both live in East Harlem and neither of their families had a statement for NBC New York.

LOS ANGELES (Reuters) ? Harold and Kumar return to theaters on Friday in "A Very Harold & Kumar 3D Christmas," the third film in the comedy franchise that includes "Harold & Kumar Go to White Castle" and "Harold & Kumar Escape from Guantanamo Bay."
This time, the dope-smoking friends -- now estranged and each with new sidekicks -- reunite to replace a prized Christmas tree Kumar accidentally burned down. While doing so, they get involved with Ukrainian mobsters, a waffle-making robot and a Christmas pageant starring Neil Patrick Harris.
Actor Kal Penn, 34, reprises his role as Kumar after taking a sabbatical to work for President Barack Obama in the White House Office of Public Engagement.
He spoke to Reuters about the film, his time away from Hollywood and what he's been doing since coming back.
Q: Why make a Harold and Kumar Christmas movie?
A: "What I love about the 'Harold & Kumar' movies is they aren't stoner movies like 'Cheech & Chong' and 'Pineapple Express.' Compared to them, Harold and Kumar aren't high for most of their movies. So I always view them as a buddy comedy first, which I think gives them liberty to do a Christmas movie, or a Guantanamo Bay movie."
Q: But Christmas movies have very specific holiday themes and don't usually involve characters like Harold and Kumar.
A: "This is a traditional Christmas movie in a lot of ways: there is a lot of heart, it is about friendship more than anything else and about a mysterious package bringing people together. Sure Harold and Kumar are obnoxious and vulgar, but it comes from such a pure-hearted place."
Q: Has President Obama seen the "Harold & Kumar" movies?
A: "I never figured out a good moment to tell the President of the United States that I was a stoner who escaped from Guantanamo Bay. When do you tell him that? Is it after he announces to the world that he took care of Bin Laden ... You know what I mean?"
Q: This is your first project since leaving Hollywood for Washington. How did you get lured back?
A: "When I left (the TV series) 'House,' I was going to do a two-year sabbatical in D.C. After I finished my first year, I got a call from the 'Harold & Kumar' producers who said, 'We're doing another movie this summer.' I said, 'I'm on a two-year sabbatical, how about next summer?' But I had a contractual obligation to do the movie when they wanted to do it. So I had to resign from my post to go shoot it."
Q: But you went back to the White House. What happened?
A: "I was only gone for two and a half, three months. When I was done, they still hadn't filled the position (in Washington) with a full-time person, so I reapplied. I told them, 'I made the commitment to serve for two years and I need to finish my additional year.' It ended up working out and I went back and finished. Now I'm acting full time."
Q: Did you leave Washington a changed man?
A: "It does change you as a person. I feel like I've become a lot more patient. Change is not a tweet. You don't just send a tweet and suddenly the world changes. It takes a lot of work, a lot of advocacy."
Q: You consider yourself an Independent, unaffiliated with either major political party. Do you think President Obama got enough done during his one term, or does he deserve another?
A: "If you look at the list of things he accomplished in just three years it's astronomical. The stuff I worked on, like the American Opportunity Tax Credit, the president increased it to $2500. That's $2500 to anyone who wanted to go to college. Then there was the Don't Ask Don't Tell repeal. But there's always more that can be done. There's immigration reform and climate change that he's still trying to advocate for, so yes, I do think he needs more time to accomplish everything."
Q: Where is the future for you, Hollywood or Washington?
A: "I'm on 'How I Met Your Mother' through the end of the year and I'm developing a workplace comedy for NBC. I'd love to do a fourth or fifth 'Harold & Kumar' if it's in the cards. I'm also currently helping with the president's reelection campaign. I hope to still do both."
